Famille très ancienne et noble, originaire de Gubbio. Au fil des siècles, divers membres de cette illustre lignée se sont répandus dans différentes régions d'Italie, où ils ont été accueillis dans des rôles importants dans la ville où ils vivaient. A Gubbio en 1163, un certain Rinaldo était consul et capitaine. Plus tard, un Michele, de Brunamonte della Serra, avec son fils Bernardo, s'installa à Bologne. En 1341 et 1347, d'autres membres de cette famille étaient à Bologne, mais le premier dont on peut parler avec certitude est Napoléon della Serra, alias della Malvasia, qui vécut en 1380, dont descendit Francesco, qui eut deux épouses: Rachele Caprara, le premier et Lucrezia Bottrigari, la seconde. Cette famille, devenue bolognaise, commença à se faire remarquer peut-être pour ses richesses.
